This reagent is available as a colourless powder which is soluble in chloroform, glycerol, benzene and less soluble in water. Chemically, Acetamide can be considered as an intermediate between acetone, which has two methyl (CH3) groups on either side of the carbonyl (CO), and urea which has two amide (NH2) groups in those locations. The addition of 5% (w/v) aqueous acetamide to a PCR mixture minimizes non-specific annealing of primers and prevents preferential amplification of replication artefacts.
