Description
Multiparameter flow cytometric analysis of CCR7 CD197 on Human peripheral blood leucocytes and lymphocytes. Human whole blood was stained with BD Horizon R718 Mouse Anti-Human CD4 Cat. No. 567092/567234 , BD Horizon BV421 Mouse Anti-Human CD8 Cat. No. 562428/562429 , PE Mouse Anti-Human CD45RA Cat. No. 555489/561883 antibodies, and with either BD Horizon BUV395 Mouse IgG1, kappa Isotype Control Cat. No. 563547 or BD Horizon BUV395 Mouse Anti-Human CCR7 CD197 antibody Cat. No. 569108 at 2.0 mug/test. Erythrocytes were lysed with BD FACS Lysing Solution Cat. No. 349202 . Flow cytometry and data analysis were performed using a BD FACS LSRFortessa X20 Flow Cytometer and FlowJo software. Data shown on this Technical Data Sheet are not lot specific. Plots 1 and 2: Bivariate pseudocolor density plots showing the correlated expression of CCR7 CD197 or Isotype control staining versus side light-scatter SSC-A signals were derived from gated events with the forward and side light-scatter characteristics of intact leucocytes. Plot 3: The two-parameter pseudocolor density plot showing the correlated expression of CCR7 CD197 versus CD45RA was derived from CD4-positive T cell gated events with the forward and side light-scatter characteristics of intact lymphocytes. Please note that we have observed considerable donor-to-donor variation in the proportion of CCR7 CD197 -negative CD4-positive lymphocytes. Plot 4: The two-parameter pseudocolor density plot showing CCR7 CD197 staining versus CD45RA was derived from CD8-positive T cell gated events with the forward and side light-scatter characteristics of intact lymphocytes.
