Description

Multicolor flow cytometric analysis of CD4 expression on mouse splenocytes. Splenic leucocytes were stained simultaneously with PE Hamster Anti-Mouse CD3e antibody Cat. No. 553064/553063/561824 and with either BD Horizon BV421 Rat IgG2a, kappa Isotype Control Cat. No. 562603; Left Panel or BD Horizon BV421 Rat Anti-Mouse CD4 antibody Cat. No. 562891; Right Panel . Two-color flow cytometric dot plots show the correlated expression patterns of CD4 or Ig Isotype control staining versus CD3 for gated events with the forward and side light-scatter characteristics of viable splenic leucocytes. Flow cytometry was performed using a BD LSR II Flow Cytometer System. Immunohistofluorescent analysis of CD4 expression by cells within C57BL/6 mouse spleen. A mouse spleen cryosection 5 mum was fixed with BD Cytofix Fixation Buffer Cat. No. 554655 , blocked with 5 % goat serum and 1 % BSA diluted in 1x PBS, and stained with BD Horizon BV421 Rat Anti-Mouse CD4 antibody Cat. No. 562891, pseudo-colored green , Alexa Fluor 647 Rat Anti-Mouse CD45R/B220 antibody Cat. No. 557683, pseudo-colored blue , and BD Horizon BV480 Rat Anti-Mouse CD31 antibody Cat.
