Description

Multiparameter flow cytometric analysis of CD25 IL-2 Receptor alpha expression on unstimulated Left and Middle Plots and stimulated Right Plot Mouse splenic leukocytes. Left and Middle Plots- BALB/c Mouse splenic leukocytes were treated with BD Pharm Lyse Lysing Buffer Cat. No. 555899 to lyse erythrocytes, washed and preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody Mouse BD Fc Block Cat No. 553141/553142 . The leukocytes were then stained with PE Rat Anti-Mouse CD4 antibody Cat. No. 553049 and with either no antibody BD Horizon BV480 Autofluorescence control; Left Plot or BD Horizon BV480 Rat Anti-Mouse CD25 IL-2 Receptor alpha antibody Cat. No. 570319/570397; Middle Plot at 1 mug/test. The bivariate pseudocolor density plots showing the correlated expression of CD25 IL-2 Receptor alpha or Ig Isotype control staining versus CD4 were derived from gated events with the forward and side light-scatter characteristics of viable splenic leukocytes. Right Plot- BALB/c Mouse splenic leukocytes were stimulated for 3 days with Concanavalin A ConA . The stimulated cells were preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody and then stained with either no antibody BD Horizon BV480 Autofluorescence control; dashed line histogram or BD Horizon BV480 Rat Anti-Mouse CD25 IL-2 Receptor alpha antibody solid line histogram at 1 mug/test. BD Via-Probe Cell Viability 7-AAD Solution Cat. No. 555815/555816 was added to cells right before analysis. The fluorescence histogram showing the CD25 IL-2 Receptor alpha or Ig Isotype control staining was derived from gated events with the forward and side light-scatter characteristics of viable 7-AAD-negative lymphoblasts. Flow cytometry and data analysis were performed using a BD LSRFortessa X-20 Cell Analyzer System and FlowJo Software. Data shown on this Technical Data Sheet are not lot specific.
