Multiparameter flow cytometric analysis of IL-1ra expression by stimulated Human peripheral blood mononuclear cells PBMC . Human PBMC were cultured with Recombinant Human IFN-gamma protein Cat. No. 554617 at 20 ng IFN-gamma/ml for 2h; 37C and then lipopolysaccharide LPS 1 mug LPS/ml for 24 h; 37C was added with Protein Transport Inhibitor BD GolgiStop Cat. No. 554724 . Cells were fixed with BD Cytofix Fixation Buffer Cat. No. 554655 and then washed and permeabilized with BD Perm/Wash Buffer Cat. No. 554723 . The cells were stained in BD Perm/Wash Buffer with either BD Horizon R718 Mouse IgG1, kappa Isotype Control Cat. No. 566928; Left Plot or BD Horizon R718 Mouse Anti-Human IL-1ra antibody Cat. No. 571484/571492; Right Plot .
