Description

Two-color flow cytometric analysis of T-bet expression in Human peripheral blood lymphocytes. Human whole blood was treated with BD Phosflow Lyse/Fix Buffer Cat. No. 558049 to lyse erythrocytes and fix leukocytes. The leukocytes were permeabilized by treatment with BD Phosflow Perm Buffer III Cat. No. 558050 . The cells were then washed and stained with BD Horizon BV421 Mouse Anti-Human CD3 antibody Cat. No. 562426 and with either BD Horizon R718 Mouse IgG1, kappa Isotype Control Cat. No. 566928; Left Plot or BD Horizon R718 Mouse Anti-T-bet antibody Cat. No. 568173/568175; Right Plot at 0.5 mug/test. The bivariate pseudocolor density plot showing the correlated expression of T-bet or Ig Isotype control staining was derived from gated events with the forward and side light-scatter characteristics of intact lymphocytes.
