Multicolor flow cytometric analysis of RORgammat expression in Mouse thymocytes. C57BL/6 Mouse thymocytes were stained with BD OptiBuild BV421 Rat Anti-Mouse CD4 Cat. No. 740007 and BD Horizon BUV395 Rat Anti-Mouse CD8a Cat. No. 563786 antibodies, and then fixed and permeabilized using the BD Pharmingen Transcription Factor Buffer Set Cat. No. 562574/562725 . The fixed and permeabilized cells were then stained with either BD Horizon RB705 Mouse IgG2a, kappa Isotype Control Cat. No. 570254 or BD Horizon RB705 Mouse Anti-Mouse RORgammat antibody Cat. No. 570259/570260 at 0.06 mug/test. The overlapping histograms showing either Ig Isotype control staining Left Panel or the expressed levels of RORgammat Right Panel were derived from either gated CD4+CD8- single-positive dashed line histograms or CD4+CD8+ double-positive solid line histograms thymocytes. The fluorescence histograms were derived from events with the forward and side light-scatter characteristics of intact cells.
