Description

Two-color flow cytometric analysis of T-bet expression in Human peripheral blood lymphocytes. Human whole blood was treated with BD Phosflow Lyse/Fix Buffer Cat. No. 558049 to lyse erythrocytes and fix leukocytes. The leukocytes were permeabilized by treatment with BD Phosflow Perm Buffer III Cat. No. 558050 . The cells were then washed and stained with BD Horizon BV421 Mouse Anti-Human CD3 antibody Cat. No. 562426 and with either BD Horizon RB705 Mouse IgG1, kappa Isotype Control Cat. No. 570261; Left Plot or BD Horizon RB705 Mouse Anti-T-bet antibody Cat. No. 570294/570295; Right Plot . The bivariate pseudocolor density plot showing the correlated expression of T-bet or Ig Isotype control staining versus CD3 was derived from gated events with the forward and side light-scatter characteristics of intact lymphocytes. Flow cytometry and data analysis were performed using a BD FACSymphony A5 Cell Analyzer System and FlowJo software.
