Flow cytometric analysis of CD83 expression on viable cultured Human dendritic cells. Human peripheral blood monocytes were treated with 20 ng/ml of Recombinant Human IL-4 Cat. No. 554605 , and 20 ng/ml of Recombinant Human GM-CSF Cat. No. 550068 proteins for 5 days. Recombinant Human TNF protein Cat. No. 554618; 20 ng/mL was then added and the cells were cultured for 2 more days at 37C. The cells were then stained with either BD Horizon RY586 Mouse IgG1, kappa Isotype Control Cat. No. 568097; dashed line histogram or with the BD Horizon RY586 Mouse Anti-Human CD83 antibody Cat. No. 569650/569651; solid line histogram . DAPI 4,6-Diamidino-2-Phenylindole, Dihydrochloride Solution Cat. No. 564907 was added to cells right before analysis. The fluorescence histogram showing CD83 expression or Ig Isotype control staining was derived from gated events with the forward and side light-scatter characteristics of viable DAPI-negative dendritic cells.
