Description
Flow cytometric analysis of Bcl-2 expression in Human peripheral blood leukocyte populations. Human peripheral blood was treated with BD FACS Lysing Solution Cat. No. 349202 to lyse erythrocytes and fix leukocytes. The leukocytes were washed with BD Pharmingen Stain Buffer FBS Cat. No. 554656 , pelleted by centrifugation and then resuspended and left in BD Phosflow Perm Buffer II Cat. No. 558052; chilled to -20C before use for 30 minutes on ice. The cells were washed twice with BD Pharmingen Stain Buffer FBS and then stained with either Alexa Fluor 488 Mouse IgG1, kappa Isotype Control Cat. No. 567121; Left Plot or Alexa Fluor 488 Mouse Anti-Human Bcl-2 antibody Cat. No. 569841/569917; Right Plot . The pseudocolor density plot showing the correlated expression of Bcl-2 or Ig Isotype control staining versus side light-scatter signals was derived from gated events with the forward and side light-scatter characteristics of intact leukocytes. Flow cytometry and data analysis were performed using a LSRFortessa Cell Analyzer System and FlowJo Software.
