Multiparameter flow cytometric analysis of IL-1ra expression in stimulated Human PBMC. Human PBMC were cultured with Recombinant Human IFN-gamma protein Cat. No. 554617 at 20 ng IFN-gamma/ml for 2h; 37C, Cat. No. 554616/554617 and then lipopolysaccharide LPS 1 mug LPS/ml for 24 h; 37C was added with Protein Transport Inhibitor BD GolgiStop Cat. No. 554724 . Cells were fixed with BD Cytofix Fixation Buffer Cat. No. 554655 and then washed and permeabilized with BD Perm/Wash Buffer Cat. No. 554723 . The cells were stained in BD Perm/Wash Buffer with either APC Mouse IgG1, kappa Isotype Control Cat. No. 554681; Left Plot or APC Mouse Anti-Human IL-1ra antibody Cat. No. 570735/570736; Right Plot . The pseudocolor density plot showing the correlated expression of IL-1ra or Ig Isotype control staining versus side-light scatter SSC-A signals was derived from gated events with the forward and side-light scatter characteristics of intact PBMC. Flow cytometry and data analysis were performed using a BD LSRFortessa Cell Analyzer System and FlowJo Software .
