Flow cytometric analysis of Human Neutrophil Elastase expression in Human peripheral blood granulocytes and tumor cell lines. Human peripheral blood granulocytes Left Panel prepared by density gradient centrifugation and cells from the Human Jurkat Acute T cell leukemia, ATCC TIB-152; Middle Panel and Human U-937 Histiocytic Lymphoma, ATCC CRL-1593, Right Panel were fixed with BD Cytofix Fixation Buffer Cat. No. 554655 , washed and then permeabilized with BD Perm/Wash Buffer Cat. No. 554723 . The cells were then stained in BD Perm/Wash Buffer with either PE Mouse IgG1, kappa Isotype Control Cat. No. 554680; Dotted Line Histograms or PE Mouse Anti-Human Neutrophil Elastase antibody Cat. No. 568908;568909; Solid Lines at 0.5 mug/test. The fluorescence histograms showing Neutrophil Elastase expression or Ig Isotype control staining were derived from gated events with the light-scatter characteristics of intact cells.
