Western Blot analysis of Sox2 in Human Neural Stem Cells NSC and Mouse Embryonic Stem ES cell line. Lysates of NSC derived from H9 human ES cells WiCell, Madison, WI, Left and ES-E14TG2a mouse ES cells ATCC CRL-1821, right were probed with Purified Mouse anti-Sox2 monoclonal antibody at concentrations of 0.125 lanes 1 , 0.06 lanes 2 , and 0.03 mug/ml lanes 3 . Human and mouse Sox2 were identified as bands of 36 kDa. Immunoflourescent staining of Sox2 in human embryonic stem ES cells. H9 human ES cells WiCell Madison, WI , passage 52 grown in mTESR1 medium StemCell Technologies on BD Matrigel hESC-qualified Matrix Cat. No. 354277 , were fixed with BD Cytofix Fixation Buffer Cat. No. 554655 , permeabilized with BD Phosflow Perm Buffer III Cat No. 558050 , and stained with Purified Mouse anti-Sox2 monoclonal antibody pseudo colored green at 10 mu g/mL. The second step reagent was Alexa Fluor 488 goat anti-mouse Ig Life Technologies , and counter-staining was with Hoechst 33342 pseudo-colored blue . The images were captured on a BD Pathway 435 Cell Analyzer and merged using BD Attovision Software. Permeabilization with BD Perm/Wash Cat No. 554723 or 0.1 % Triton X-100 buffer is also suitable for use with this antibody.
