Description

Flow cytometric analysis of Stat5 PY694 expression in IL-2-stimulated Human CD4+ T lymphocytes. Human peripheral blood mononuclear cells were cultured with 100 ng/mL recombinant Human IL-2 protein Cat. No. 554603 for 15 minutes at 37C. The cells were then fixed with BD Phosflow Fix Buffer I Cat. No. 557870 , washed with Stain Buffer FBS Cat. No. 554656 , and then permeabilized 30 min on ice with chilled BD Phosflow Perm Buffer III -20C prior to use; No. 558050 . The cells were washed twice and stained in Stain Buffer FBS with either BD Horizon RY775 Mouse IgG1, kappa Isotype Control Cat. No. 571408; dashed line histogram or BD Phosflow RY775 Mouse Anti-Stat5 pY694 antibody Cat. No. 571624/571706; solid line histogram . The histogram showing Stat5 pY694 expression or Ig Isotype control staining was derived from gated events with the forward and side light-scatter characteristics of intact lymphocytes.
