Immortalized Mouse MyD88 -/- Hepatic Stellate Cells

Description

The Immortalized Mouse MyD88/ Hepatic Stellate Cells were derived from MyD88 knockout mice, generated via targeted disruption of the MyD88 gene in embryonic stem cells using electroporation of a linearized targeting construct. Primary hepatic stellate cells were isolated by enzymatic digestion and Percoll density gradient centrifugation, then immortalized through Fugene-mediated transfection of SV40 Large T antigen pCMV-SV40 . As MyD88 is a critical adaptor protein downstream of TLR4 signaling, these cells show minimal NF-kappaB activation in response to LPS, even when expressing wild-type TLR4. They are ideal for studies of liver fibrosis, TLR4-mediated inflammatory signaling in scar formation, and apoptosis resistance linked to TLR4 activation.
