Construction Of Recombinant DNA Molecule Is Dependent On The Ability To Covalently Seal Single Stranded Nicks In The DNA.This Process Is Accomplished Both In Vivo And In Vitro By The Enzyme DNA Ligase.This Enzyme Catalyzes The Formation Of Phosphodiester Bonds Between Juxtaposed 5'Phosphate And A3'Hydroxyl Terminus Of Double Stranded DNA. It Can Repair Single Stranded Nicks In Double Stranded Dna And Join Double Stranded DNA Restriction Fragments Having Either Bluntend So Homologous Cohesive Ends.
