Description
pBI-CMV2 is a mammalian bidirectional expression vector designed to constitutively express a protein of interest and AcGFP1, a green fluorescent protein derived fromAequorea coerulescens. The vector allows straightforward detection of transfected mammalian cells by flow cytometry or fluorescence microscopy, as cells expressing the protein of interest can be quickly identified by screening for AcGFP1 fluorescence. Protein expression is driven by one of two constitutively active human cytomegalovirus promoters:PCMV IEdrives the expression of the protein of interest, andPminCMVdrives the expression of AcGFP1. Back Required Products Linear Hygromycin Marker is a short ds DNA fragment consisting of the hygromycin resistance gene regulated by the SV40 promoter and ending with the SV40 polyadenylation signal. The 2.1-kb marker is compatible with any expression vector including Tet System vectors for generating stable transfectants using CalPhos transfection reagent or a liposome-mediated transfection protocol. Back Back Cotransfection of a hygromycin or puromycin linear selection marker achieves a higher number of positive clones than using either a single response vector containing a selection marker, or cotransfecting with a circular selection marker. Linear Puromycin Marker is a short ds DNA fragment consisting of the puromycin resistance gene regulated by the SV40 promoter and ending with the SV40 poly-adenylation signal. The 1.8-kb marker is compatible with any expression vector including Tet System vectors for generating stable transfectants using CalPhostransfection reagent or a liposome-mediated transfection protocol. Back Cotransfection of a hygromycin or puromycin linear selection marker achieves a higher number of positive clones than using either a single response vector containing a selection marker, or cotransfecting with a circular selection marker.
